Great Build
After accompanying Reed Richards, Benjamin Grimm and her brother Johnny on a trip to space in an experimental rocket that Reed had built, the group was bathed in cosmic rays leading to the development of amazing powers. Sue's powers manifested in the form of light-bending force fields that allow her to turn invisible, protect herself and others, as well as use them as a destructive force to defend herself.Marvel Source
She is informally known as the most powerful member of the Fantastic Four due to the limitless potential of her powers.
I just revised the build. Not much has changed as it still works great, but I updated and expanded it a bit.

Introduction
NOTE: Sorry I cannot update this build with Sue's latest power set until MarvelBase makes it available.Sue is my favorite hero not just because she's extremely capable, but because I find her so much fun to play and fun to watch. While most other heroes have something I don't like, I like everything about Sue. I find it a joy to run with her through all the level 60 content and she's the only one I've felt motivated to prestige.
I will admit however that I have not raided and I'd definitely defer to an experienced raider for the best raiding build.
Powers
Dynamic Force Powers(MAX)
Not the fastest trash clearer, but I never tire of watching the animation. If you have to take out the trash, you may as well have fun doing it, right? You can target pretty much anything handy and Bouncing Discs seek out all the baddies for you. But if you can, target a blue or yellow since they last longer. To use it to greatest effect, run around until you’re in range of multiple targets. It’s pathetic against single targets, but just find a couple more enemies and it destroys them. Like Fezzik, Sue specializes in groups.

Max this to layer significant damage on the more stationary targets. As with all ground effects it can be frustrating with highly mobile bosses, but the area is good and you can just recast as much as you like since you've got plenty of power to burn. Plus it looks really cool. While large in scale, her transparent animations don't obscure the entire screen.

This is your one offensive passive and you have plenty of points to max it out, so go ahead! Note that I pretty much just ignore the Focus resource but this power is worthwhile even without the Focus bonus.

I don't like "1 point wonder" powers, especially in my Signature skill. If it's worth a place on my hotbar it should be worth investing in. This signature delivers great aoe damage every 30 seconds. Pop it when a mob of mobs gets too up close and personal for comfort. Plus it gives you nice situational flexibility. Activate once for some big aoe damage and a damage negation shield, or repeat to blow the shields for added carnage and knockback.
Invisibility Powers

I consider this the defining Invisible Woman power. It makes you feel that you are truly the Invisible Woman. You are able to run around safely, turning invisible almost absent-mindedly as Sue would. Toggle it on and leave it on. I keep it on my secondary hotbar and forget about it after toggling it on.

Spam Resonating Wave whenever you can for spirit plus some bonus damage with a stun. And don’t worry whether you have any mobs in range. Just hit it regardless for the spirit. You can also use it as an opener while invisible for the damage bonus. Many weaker mobs will fall and go boom before they even know you're standing in their midst.

This is your deceptively potent big damage spirit spender. You should have plenty of spirit with Bouncing Discs doing most of the work, so feel free to spam this whenever the mood strikes you, particularly against nasty bosses. And like Bouncing Discs, Seeker Orbs are an intelligent targeting power allowing you to just hit it and trust them to seek out the bad guys.

This is one of the best passives in the game. Defense, deflection, and major regeneration on hit, plus an invisibility save. This will keep you tossing those energy frisbees long after you should have face-planted. You’d be crazy not to max this!

Despite the great buffs, I honestly don't normally bother with this. You really don't need the spirit and should have few survivability issues. And note that the Unaware Target bonuses don't apply unless the mob is actively targeting someone else, so they do little good on solo boss encounters where you could use it most. Still, feel free to spam it for an additional edge.

I don't even keep this on my hotbar, but feel free to include it for quick escapes from tight situations.
Ultimate
Spheroid Typhoon is your incredible “run around and destroy everything every 10 minutes” power. It's devastating not only against massive numbers in Midtown, but essential to bring down bosses quickly. And more importantly, the huge force bubble is one of the coolest animations in the game. Fly around with your big sphere up for the ultimate bubble fun! Hers is an ultimate that I feel is worth investing in with as many upgrades as you can manage.
Play Style
While invisible, run into a crowd and hit Resonating Aura. Target the hardest mob, start tossing Bouncing Discs and don't stop. Lay down a Storm of Spheres under groups, elites, or bosses. Fire off Seeker Orbs as quick as you can to drop elites and bosses. Hit Quantum Compression if you find yourself surrounded, and keep firing it to clear the area around you. If you’re in fear for your life (which seriously seldom happens), go invisible or blitz out of danger. Oh, and don't forget to use your Ultimate as much as you can to destroy large crowds or tear apart bosses. The challenge is keeping those hard-hitting bosses within your Typhoon.Run around, not to avoid, but to gather additional enemies. This makes for a very fast, fun, and aggressive play style.
Did the doorbell just ring? Is it the pizza guy with that pepperoni special? No problem! Just pause a moment, let Sue turn invisible wherever she is, and go enjoy. She'll still be just fine whenever you get back. (Not really all THAT fantastic, are ya Reed?)
Gear
Find everything you can that boosts Dynamic Force powers first, and Invisibility powers second.Your other gearing choices are pretty easy with Sue. I pretty much go for all offensive stats that give me the most energy, ranged, and area damage boosts.
For her Artifacts, it's hard to go too wrong, but I like the Advanced Kree Hyper-Optics for the slow proc. The Sharkstone Tooth can give you extra health on hit if you need it. In fact, anything with a ‘to hit’ proc is a good candidate for Sue since she hits so quickly. I have a number of "life on hit" effects and I very rarely use a medkit. Hulkbuster Munitions are the only less than obvious choice because Sue can use the extra firepower against bosses. The knockback effect is welcome for her as well.
For her Medallion you have also many good choices. But as a default, Doctor Octopus is a no-brainer to increase the radius and damage of your fantastic AEs. Don't feel too cliche using it. For the more defensive-minded, the Blob medallion is worth considering. In addition to all the defensive buffs and another slow on hit chance, you also get bonus damage rating for aware targets.
For her Legendary, you have the usual choices but I went with the Cosmic Control Rod partly because it makes me feel more in character, and I’m quite happy with it. The only thing I lament about the CCR is the wasted spirit buff. It's totally unnecessary with this build as Sue stays at max spirit no matter how much you spam her spenders. Don't bother gearing for more spirit as you won't need it.
For her Relic, I used to use Lemuria for health on hit. But really I have such overkill on health that I more recently changed to Wakanda for the added DOT rating applied to Storm of Spheres and Spheroid Typhoon. I have not missed the health at all.
For her Core, feel free to go with purely offensive buffs, or defense if you're weak there for some reason, or even health on hit if you need it. Bottom line, use these to tweak your particular situation.

Wrap Up
That’s about it! It’s a simple build but not simple-minded. It allows for a lot of spontaneity and doesn’t feel at all mechanical and repetitive. You'll never have to worry about spirit or your secondary resource bar, and honestly seldom even worry about watching your health.Have fun playing with the Disco Queen!
